چکیده

Traditional approaches to pathological diagnosis include macroscopic examination and formalin fixation with the preparation of paraffin sections stained with a variety of histochemical techniques. These routine approaches however may be disappointing in the assessment of many conditions and occasionally fail to render a precise diagnosis or provide adequate prognostic information. Consequently there has been growing interest within the histopathological community in the state of the nucleus, DNA and proliferation markers. One of the most recent produces of this interst is the study of Nucleolar Organiser Regions (NORs) using a simple silver reduction technique (AgNOR method). Nucleolar organiser regions are an essential part of the machinery of the nucleolus (Howell, 1982; Derenzini et al., 1983). They can be seen as electron lucent areas at the ultrastructural level (fibrillar centres) which are the interphase equivalent of the condensed chromosomal NORs (Jordan & McGovern, 1981; Hernandez-Verdun, 1986). These structures can be seen at the light microscope level using a variety of techniques including the AgNOR method (Hsu et al., 1975; Ploton et al., 1986). They are involved in ribo-

Nucleolar organiser regions: new prognostic variable in breast carcinomas.

Nucleolar organiser regions (NORs), which are important for regulating protein synthesis, were identified in 20 breast carcinomas by means of a silver (Ag) staining technique. Infiltrating neoplasms with metastases in four or more axillary lymph nodes possessed, on average, a greater number of AgNORs per cell nucleus compared with neoplasms without nodal disease, or with one to three positive l...

Proliferating cell nuclear antigen (PCNA) and nucleolar organiser regions in Hodgkin's disease: correlation with morphology.

AIM To define the distribution of proliferating cell nuclear antigen (PCNA) and silver staining nucleolar organiser regions (AgNORs) in Hodgkin's disease. METHODS PCNA was shown in a series of 34 cases of Hodgkin's disease using immunohistochemical methods. In a second series of 46 cases the AgNOR technique for interphase nucleolar organiser regions was studied. Both series comprised routinel...

Improved silver staining of nucleolar organiser regions in paraffin wax sections using an inverted incubation technique.

A new simple modification to the silver staining of nucleolar organiser regions (AgNORs) was devised which, by performing the incubation with the slide inverted, results in minimal undesirable background staining, a persistent problem. Inverted incubation is facilitated by the use of a commercially available plastic coverplate. This technique has several additional advantages over other publish...

Differentiation of malignant from normal and reactive mesothelial cells by the argyrophil technique for nucleolar organiser region associated proteins.

Nucleolar organiser regions represent nucleolar activity in both normal and malignant cells. More numerous nucleolar organiser regions have been found in cells from many forms of malignancy than in normal cells. An argyrophilic method of staining these regions (the AgNOR technique) has been applied to specimens of pleura obtained by biopsy or at necropsy in an attempt to differentiate between n...

Nucleolar organiser region associated proteins in cutaneous melanotic lesions: a quantitative study.

Using a silver staining technique, nucleolar organiser regions were identified in routinely processed paraffin sections of a range of dermal melanotic lesions. The technique shows argyrophilic NOR associated proteins (AgNORs), which are seen in nuclei as black dots. Although the nuclei of melanocarcinoma in situ, melanocarcinoma per se, and lentigo maligna contained similar numbers of AgNORs, i...
